Lacking any intrinsic enzymatic activity, CrkL is composed of one SH2 domain a n d two SH3 domains (1). The SH2 domain of CrkL as been shown to interact with c-Cbl, Cas, Hef-1 a n d Paxillin (2); while the N-terminal SH3 can bind to c-ABL, Sos1 a n d C3G (3). Additionally, the phosph o r ylated f o r m of CrkL (pY207) binds to BCr-ABL oncogene in chronic myelogenous leukemia cells (4).
