Synonyms rHuSignal regulatory protein α, Fc Chimera; CD172a; SHPS-1; BIT; MYD1 ; 重组人信号调节蛋白α, Fc 嵌合体 (HEK293 细胞表达) Species HumanSource HEK 293 Accession P78324 Gene ID 140885 Molecular Weight 85-100 kDa AA Sequence EEELQVIQPD KSVLVAAGET ATLRCTATSL IPVGPIQWFR GAGPGRELIY NQKEGHFPRV TTVSDLTKRN NMDFSIRIGN ITPADAGTYY CVKFRKGSPD DVEFKSGAGT ELSVRAKPSA PVVSGPAARA TPQHTVSFTC ESHGFSPRDI TLKWFKNGNE LSDFQTNVDP VGESVSYSIH STAKVVLTRE DVHSQVICEV AHVTLQGDPL RGTANLSETI RVPPTLEVTQ QPVRAENQVN VTCQVRKFYP QRLQLTWLEN GNVSRTETAS TVTENKDGTY NWMSWLLVNV SAHRDDVKLT CQVEHDGQPA VSKSHDLKVS AHPKEQGSNT AAENTGSNER Biological Activity 2 µg/mL (100 µL/well) of immoblized recombinant human CD47-His can bind human Signal regulatory protein alpha-Fc with a linear range of 0.25-185 ng/mL. Appearance Lyophilized powder. Formulation Lyophilized after extensive dialysis against PBS, 5% trehalose and mannitol. Endotoxin Level <0.2 EU/μg, determined by LAL method. Reconstitution Reconstitute the lyophilized recombinant Human Signal regulatory protein alpha, Fc Chimera (HEK293-expressed) (rHuSignal regulatory protein α, Fc Chimera) to 100 µg/mL using ddH2O or diluted with PBS. Storage & Stability Lyophilized recombinant Human Signal regulatory protein alpha, Fc Chimera (HEK293-expressed) (rHuSignal regulatory protein α, Fc Chimera) is stored at -20°C. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer. It is recommended to freeze aliquots at -20°C or -80°C for extended storage. Shipping Room temperature in continental US; may vary elsewhere. Background Signal regulatory protein α (SIRPα) is a glycoprotein receptor that recruits and signals via the tyrosine phosphatases SHP-1 and SHP-2.The cytoplasmic domain of SIRPα contains four immunoreceptor tyrosine-based inhibition motifs (ITIMs), which upon phosphorylation recruit and activate SH2-domain-containing phosphotyrosine phosphatases (PTPase) SHP-1 and SHP-2. In macrophages SIRPα can negatively regulate the phagocytosis of host cells and the production of tumor necrosis factor alpha. SIRPα ligation induces macrophage NO production through the cooperative action of JAK/STAT and PI3-K/Rac1/NOX/H2O2 signaling pathways. It proposes that SIRPα may function as an inhibitory receptor[1]. |
