Recombinant Mouse Thrombopoietin, His (HEK293-expressed) 是一种在 HEK293 细胞中产生的带有标签的重组小鼠血小板生成素 (TPO),在 HEK293 细胞中产生。Recombinant Mouse Thrombopoietin 是巨核细胞生长和分化的主要调节因子。 Synonyms rMuThpo, His; C-mpl ligand; MGDF ; 重组小鼠血小板生成素,His 标签 (HEK293 细胞表达) Species MouseSource HEK 293 Accession P40226 Gene ID 21832 Molecular Weight 62-70 kDa AA Sequence SPVAPACDPR LLNKLLRDSH LLHSRLSQCP DVDPLSIPVL LPAVDFSLGE WKTQTEQSKA QDILGAVSLL LEGVMAARGQ LEPSCLSSLL GQLSGQVRLL LGALQGLLGT QLPLQGRTTA HKDPNALFLS LQQLLRGKVR FLLLVEGPTL CVRRTLPTTA VPSSTSQLLT LNKFPNRTSG LLETNFSVTA RTAGPGLLSR LQGFRVKITP GQLNQTSRSP VQISGYLNRT HGPVNGTHGL FAGTSLQTLE ASDISPGAFN KGSLAFNLQG GLPPSPSLAP DGHTPFPPSP ALPTTHGSPP QLHPLFPDPS TTMPNSTAPH PVTMYPHPRN LSQETHHHHH H Biological Activity The ED50 is <2 ng/mL as measured by MO7e cells. Appearance Lyophilized powder. Formulation Lyophilized after extensive dialysis against PBS. Endotoxin Level <0.2 EU/μg, determined by LAL method. Reconstitution Reconstitute the lyophilized recombinant Mouse Thrombopoietin, His (HEK293-expressed) (rMuThpo, His) to 100 µg/mL using ddH2O or diluted with PBS. Storage & Stability Lyophilized recombinant Mouse Thrombopoietin, His (HEK293-expressed) (rMuThpo, His) is stored at -20°C. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer. It is recommended to freeze aliquots at -20°C or -80°C for extended storage. Shipping Room temperature in continental US; may vary elsewhere. Background Thrombopoietin (TPO) is the master cytokine regulator of megakaryopoiesis. In addition to regulation of megakaryocyte and platelet number, TPO is important for maintaining proper hematopoietic stem cell (HSC) function. TPO produced mainly in the liver, stimulates both megakaryocyte colony formation and enhances megakaryocyte maturation in the bone marrow leading to increased platelet production. TPO binds to its receptor, TPOR/MPL, which is expressed on hematopoietic stem cells (HSCs), megakaryocytes and platelets. Circulating TPO concentrations are controlled by MPL via receptor-mediated internalization and degradation, and in part by sensing of senescent platelets by the Ashwell-Morell receptor in the liver[1]. |
